At about 9.15, police were called to an address on Pipers Creek Rd after reports of a stabbing.
Detective Chief Inspector Kim Fehon told the Argus that a 54-year-old man was located deceased inside the residence.
Some 20 minutes later, emergency services were called to a single vehicle accident about 300 metres away from the house where the deceased was found.
Officers freed a local man - aged 40 - from the vehicle. He suffered non-life threatening injuries and was taken under police guard to hospital.
Insp Fehon said the deceased's injuries were consistent with being stabbed - and investigators are awaiting post-mortem results.
She said officers had conducted extensive searches of the property and Pipers Creek Rd, but the weapon had not been located.
Other people at the residence had alerted police to the incident, calling Triple Zero to report a stabbing.
Insp Fehon told the Argus that the deceased and the man in the vehicle accident were known to one another.
While there were a number of witnesses to the incident, she said anyone who may have been in the area at the time should contact police as their information could prove useful.
Detectives from Mid North Coast Police District continue with inquiries into the incident.
The 40-year-old man was released from hospital yesterday, and taken to Port Macquarie Police Station, where he was charged with murder.
He has been refused bail to appear at Kempsey Local Court later today.
Anyone with information about this incident is urged to contact Crime Stoppers: 1800 333-000 or https://nsw.crimestoppers.com.au.
