Manning River won the toss and sent Macleay Valley to the crease. This decision proved fruitful as Macleay was dismissed for 94 in the 27th over.
Manning River had their work cut out for them in the reply, losing seven wickets before reaching the target in the 23rd over.
In the round's other fixture, Hastings River defeated Gloucester at Oxley Oval in Port Macquarie.
Action continues this Sunday when Gloucester hosts Macleay Valley at Gloucester District Park and Manning River takes on Hastings River at Chatham Park.
The final will be held on February 23 at a venue yet to be determined.
Meanwhile, Premier League, Manning second grade and third grade cricket returns on Saturday.
The Premier League sides will go head to head in the final T20 matches of the season.
In second grade, Great Lakes plays Taree West at Tuncurry Oval and competition leaders Gloucester Bushmen take on Wingham. Old Bar has the bye.
Third grade matches will be Old Bar against Great Lakes, Wingham White taking on Bulahdelah, United host Old Bar Harrington and Taree West playing Wingham Blue.
