It comes after another four cases were announced overnight to 8pm October 19 bringing Kempsey's positive COVID numbers in the last two weeks to 18.
Three of the cases are linked to known cases from the Kempsey LGA and the source of the infection of the other case is under investigation.
There have been 57 cases reported in the MNCLHD since the beginning of the current NSW outbreak.
"We have been advised that two members of the school community have tested positive to COVID-19," the school issued in a statement to parents on Wednesday.
"As a result, KAS will be closed effective immediately to allow for a deep clean of the site."
All staff and students will now be required to self-isolate until they receive further advice. The school will remained closed until Friday when the situation will be reviewed.
Kempsey West Public School also shut the gates late Tuesday after it was notified of a positive COVID case in the school community.
NSW Health has urged anyone who has been unwell or has symptoms such as a fever, cough, sore throat, shortness of breath, runny nose, loss of smell/taste, muscle/joint pains, diarrhoea, nausea/vomiting or extreme tiredness to be tested at one of the COVID-19 testing clinics.
A full list of clinic locations and opening hours are listed on the NSW government website at www.nsw.gov.au/covid-19/how-to-protect-yourself-and-others/clinics.
Find available vaccination bookings at the Australian government's COVID-19 Vaccine Clinic Finder (previously Eligibility Checker), or you can also call Health Direct on 1800 571 155 for assistance to book.
